ABSTRACT

OBJECTIVES@#To evaluate the composition of bioactive substances and the antioxidant effects of jujube fruit under gut microbiota fermentation (GMF), and the inhibitory effect on cytotoxicity caused by ethyl carbamate (EC).@*METHODS@#Changes in the contents of flavonoids, polyphenols, total sugars, and reducing sugars of jujube fruit after GMF (0, 2, 6, 12, 24, and 48 h) were determined. The oxidation resistance of fermented jujube fruits (from 0 to 48 h fermentation) was evaluated using in vitro 2,2'-azinobis-(3-ethylbenzthiazoline-6-sulphonate) (ABTS) and ferric reducing antioxidant power (FRAP) assays. Inhibitory effects of 48 h-fermented jujube fruit at various concentrations (0.25, 0.50, 1.00, and 2.00 mg/mL) on EC-treated toxicity and DNA damage of Caco-2 cells were estimated using 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) and nuclear staining assays, respectively. Effects of different concentrations of jujube fruit on EC-treated Caco-2 cells' intracellular reactive oxygen species (ROS), glutathione (GSH) levels, and mitochondrial membrane potential (MMP) were also evaluated.@*RESULTS@#Jujube fruit has rich bioactive components after GMF and shows strong antioxidant capacity. Fermented jujube fruit can inhibit the cytotoxicity and DNA damage of Caco-2 cells caused by EC and reduce intracellular ROS generation, as well as restoring GSH and MMP.@*CONCLUSIONS@#Fermented jujube fruit extracts produced by GMF still contain biologically active substances which retain biological activity and antioxidation capabilities.

ABSTRACT

ABSTRACT This study was done to investigate the synergistic impacts hydro extract of jujube fruit in combination with Mesalazine (orally) and Asacol (intra-colonic) administration in ameliorating animal model of ulcerative colitis (UC). After the induction of UC and with the development of signs, the treatment groups daily received the hydro extract of jujube fruit (200 mg/kg, orally, enema), Mesalazine (30 mg/kg, orally) and Asacol (10 mg/kg, enema). After 10 days, rats were euthanized and were studied. Findings indicated a significant increase in Myeloperoxidase (161.66 ± 10.40), Nitric oxide (216.01 ± 17.55), IL-6 (138.54 ± 7.02), and TNF-α (123.87 ± 9.80) colon tissue levels and pathological damage of positive control group compared with the negative control group. Hydro extract of jujube fruit in combination with Mesalazine (orally) and Asacol (intra-colonic) group represented a higher capability in significantly decreasing Myeloperoxidase (73.33 ± 9.07), Nitric oxide (81.66 ± 10.50), IL-6 (51.69 ± 5.19), TNF-α (30.59 ± 5.50) levels and pathological damage in compared with the other treatment groups. Considering accessibility and affordability of jujube fruit and the side effects of routine drugs, taking a combination of jujube fruit with low doses of routine pharmaceutical drugs can improve and cure ulcerative colitis disease.


RESUMO Este estudo foi realizado para investigar os impactos sinérgicos do extrato aquoso do fruto da jujuba em combinação com a administração de Mesalazina (por via oral) e Asacol (intracolônico) na melhora do modelo animal de colite ulcerativa. Após a indução da colite ulcerativa e com o desenvolvimento de sinais, os grupos de tratamento receberam diariamente o extrato aquoso do fruto da jujuba (200 mg/kg, via oral, enema), Mesalazina (30 mg/kg, via oral) e Asacol (10 mg/kg, enema). Após 10 dias, os ratos foram eutanasiados e estudados. Os achados indicaram um aumento significativo dos níveis de mieloperoxidase (161,66 ± 10,40), óxido nítrico (216,01 ± 17,55), IL-6 (138,54 ± 7,02) e TNF-α (123,87 ± 9,80) no tecido do cólon e dano patológico do grupo controle positivo comparado com o grupo controle negativo. O extrato aquoso da fruta de jujuba em combinação com Mesalazina (oral) e Asacol (intracolônico) representou maior capacidade de redução significativa dos níveis de mieloperoxidase (73,33 ± 9,07), óxido nítrico (81,66 ± 10,50), IL-6 (51,69 ± 5,19), TNF-α (30,59 ± 5,50) e dano patológico em comparação com os outros grupos de tratamento. Considerando a acessibilidade e disponibilidade do fruto da jujuba e dos efeitos colaterais dos medicamentos de rotina, tomar uma combinação de jujuba com baixas doses de medicamentos farmacêuticos de rotina pode melhorar e curar a colite ulcerativa.

ABSTRACT

Objective To investigate the breeding of Carpoglyphus lactis in stored jujube from Anhui Province, so as to provide the evidence for preventing the harm of C. lactis to stored jujube. Methods The jujube samples stored for more than 6 months were collected from dried fruit shops and/or Chinese herbal medicine warehouses, which were chosen as survey sites in Anhui Province. The mites were isolated and identified under a microscope. Results Totally 19 samples infested with C. lactis were obtained from 300 samples in 17 cities of Anhui Province, the breeding rate and breeding density of C. lactis were 6.33% and 6.52 per gram respectively. The constituent ratios of different developmental stages were adult 85.72%, larva 12.27%, dormancy body 0.56%, and egg 1.45%, respectively. Conclusion The breeding density of C. lactis in the stored jujube is high and the species diversity is rich, so the control of C. lactis and the prevention of human acariasis should be strengthened.

ABSTRACT

Abstract The aim of this study was to investigate whether anaerobic metabolites could induce volatile compounds and improve aroma of dried jujube (Ziziphus jujuba Miller). Jujube fruits were incubated in a polyvinyl chloride bag containing 5% CO2 and 95% N2 for up to 168 h at 25 °C and 3 samples were randomly removed every 6 h and oven dried to a moisture content of ≅ 20%. The volatile compounds of control and 5% CO2-pretreated Chinese jujube fruits were extracted by simultaneous distillation extraction and identified by gas chromatography-mass spectrometry(GC-MS). The acetaldehyde and ethanol contents were determined by gas chromatography (GC). The results indicated that a large accumulation of acetaldehyde and ethanol caused changes in aroma composition of dried jujube products and 5% CO2 pretreatment led to an increase in the levels of some compounds, particularly esters, acetaldehydes, and ethanol, whereas the amount of acids were decreased significantly. Principal component analysis showed that integrative scores of 5% CO2 pretreatment at 120 h were the highest, and aroma quality was better than that of the control. Relatively low concentrations of anaerobic respiration metabolites are good for jujube fruit aroma composition.

ABSTRACT

<p><b>OBJECTIVE</b>To assess the biological effects of the six-herb mixture Anti-Insomia Formula (AIF) extract using caffeine-induced insomnia Drosophila model and short-sleep mutants.</p><p><b>METHODS</b>Caffeineinduced insomnia wild-type Drosophila and short-sleep mutant flies minisleep (mns) and Hyperkinetic(Y) (Hk(Y)) were used to assess the hypnotic effects of the AIF in vivo. The night time activity, the amount of night time sleep and the number of sleep bouts were determined using Drosophila activity monitoring system. Sleep was defined as any period of uninterrupted behavioral immobility (0 count per minute) lasting > 5 min. Night time sleep was calculated by summing up the sleep time in the dark period. Number of sleep bouts was calculated by counting the number of sleep episodes in the dark period.</p><p><b>RESULTS</b>AIF at the dosage of 50 mg/mL, effectively attenuated caffeine-induced wakefulness (P<0.01) in wild-type Canton-S flies as indicated by the reduction of the sleep bouts, night time activities and increase of the amount of night time sleep. AIF also significantly reduced sleeping time of short-sleep Hk(Y) mutant flies (P<0.01). However, AIF did not produce similar effect in mns mutants.</p><p><b>CONCLUSION</b>AIF might be able to rescue the abnormal condition caused by mutated modulatory subunit of the tetrameric potassium channel, but not rescuing the abnormal nerve firing caused by Shaker gene mutation. This study provides the scientific evidence to support the use of AIF in Chinese medicine for promoting sleep quality in insomnia.</p>

ABSTRACT

Objective To detect the main nutritional components of novel jujube beverage from Laoling Zizyphus Jujuba cv.Jinsixiaozao.Methods Free monosaccharides in novel jujube juice were determined by the HPLC-RID method while water-soluble polysaccharides were analyzed by UV spectrophotometric method with the phenol-sulfuric acid derivatization.Amino acid and mineral contents were obtained by the automatic amino acid analyzer and atomic absorption photometric methods, respectively.The quantification of total phenolic and flavonoids was accomplished by the UV spectrophotometric method after the respective complexation of the Folin-Ciocalteu reagent and aluminum nitrate.The contents of rutin and cyclic nucleotides were determined by the HPLC-UV method.Results The average contents of sucrose, glucose, fructose and polysaccharides in novel jujube juice were 3.85, 32.5,27.3 and 3.6 mg/mL, respectively.The total amino acid average content was 269.7 μg/mL.The mineral average contents of Ca, Fe, Mg, Zn, Mn, Cr, and Cu were 35.1,0.85, 55.8, 0.85, 0.44, 0.41, 0.21μg/mL, respectively.The novel jujube juice contained average 1.34μg/mL rutin equivalents of total phenolics and average 157.3 μg/mL gallic acid equivalents of total flavonoids.Finally, the average amounts of rutin, cAMP and cGMP were 7.15, 19.17 , and 8.11 μg/mL, respectively.Conclusion The newly developed jujube juice is worthwhile further marketing development attributed to its rich nutritional components and healthy values .

ABSTRACT

Objective: To initially establish HPLC fingerprints for analysis of 17 batches of the introdued jujube (Ziziphus jujuba) varieties in order to lay the foundation for scientific evaluation and production quality control of jujube. Methods: TOSOH TSKgel ODS-100V C18 column (150 mm × 4.6 mm, 3 μm) was used. The mobile phase was as follows: methanol and 0.3% KH2PO4 in gradient elution mode. The detective wavelength was 260 nm, the flow rate was 1.0 mL/min, and the separation was performed at 40 ℃. Similarity Evaluation System for Chromatographic Fingerprint of Chinese Materia Medica 2004A published by the state Pharmacopeia Committee of China was adopted for the fingerprint analysis on the 17 batches of introdued jujube varieties in Germplasm Nursery of Tarim University. Results: The HPLC characteristic fingerprint of jujube has been established. A total of 22 common peaks were characterized, the peaks appeared uniform and were well-separated. The uracil, hypoxanthine, adenine, cytidine, uridine, cAMP, and cGMP were determined by HPLC method. Conclusion: HPLC chromatographic fingerprint analysis technology can be used for the identification of jujube and its quality control.

Methods: By using standard methods samples were evaluated for their antimicrobial properties including additive effect of starch and non-peroxidase activity, antioxidative properties (phenol contents, flavonoid contents, 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity). Prior to this evaluation, complete physico-chemical properties including pH, color, ash contents, protein contents, moisture contents, hydroxymethyl furfural contents, total sugar contents, reducing sugar and non-reducing sugar contents were analyzed. Results: Relatively higher ash contents were found in the Siddar honey i.e. (0.590 0±0.033 6)%and small honey showed relatively higher protein contents i.e. (777.598±9.880) mg/kg. The moisture contents of tested honey samples ranged between 13.8%-16.6%, total sugar contents from 61.672%-72.420%and non-reducing sugar contents from 1.95%-3.93%. Presences of phenolic contents indicate higher antioxidant potential of these honey samples. All bacteria showed clear inhibition zones in response to tested honey samples whereas fungi and yeast showed inhibition at higher concentrations of these honey samples. For Escherichia coli, Bacillus subtilis, Salmonella typhi, Pseudomonas aeroginosa and Aspergillus niger, overall the small honey showed the higher activity than other honey samples. ABSTRACT

Objective To observe the effects of wild jujube seed decoction (WJSD) on learning memory and the levels of malondialdehyde (MDA) and the activities of supemxide dismutase (SOD) and nitric oxide synthase (NOS) in the brain of the pathogeny model rats with sleep deprivation (SD). Methods Levels of learning memory and MDA, and activities of SOD and NOS in rats' brain were assayed after SD induced by made-self multiple platform method ( MMPM), senescence induced by D-galactose, and Yin and blood-deficiency induced by eyclophosphamide and hydrocortisonum. Results After 48 h SD, the levels of learning memory was lower in the model group( ( 108.9 ± 12.5 ) s, ( 89 ± 11.5 ) s, ( 0 ) ) than those in the environmental control group ( ( 47.3 ±4.6)s,(9±1.4)s,(6.5 ±1.2))(( t=4.36,3.18,2.07, P<0.01==. While MDA, and activities of SOD and NO in rats'brain higher in the model group( (3.8 ±0.6) ,(3.0 ±0.5)nmol · mgprot-1 ,(229.7 ±25.8) ,(236.3± 25.2 ) U · mgprot - 1, ( 5.7 ± 0. 8 ), ( 5.4 ± 0.9 ) U · mgprot - 1 ) than those in the environmental control group ( (2. 1 ±0.4) ,(1.6 ±0.4)nmol · mgprot-1 ,(155.5 ±10.6) ,( 147.2 ±26.1 )U · mgprot-1 ,(2.8 ±0.7),(2.9 ± 0.5 ) U · mgprot -1 ) ( t = 2.89,3.01,6.78,5.94,3.10,3.46, P < 0.05 =. However, the observation of the groups treated with WJSD, the levels of learning memory showed a tendency in returning to normal level (P < 0.05 = ,and MDA, and activities of SOD and NO of the high dose of WJSD showed a tendency in returning to normal leve1 (P < 0.05 = ,and the low dose of WJSD showed a tendency in returning to normal level too(P < 0.05 =. Conclusion WJSD can improve the disability of learning and memory of the pathogeny rats model, and its one of mechanism maybe involve the reduction of neural cell damage with free radical and NO.

ABSTRACT

Objective: To explore the protective effects of jujube polysaccharide on liver injury model mice induced by ethionine and paracetamol. Methods: Liver injury mice model was induced by gavage with ethionine suspension or intraperitoneal injection of parcharide. ALT levels in serum and liver histopathological changes were measured, the protective effect of jujube polysaccharide on liver injury model mice induced by ethionine or paracetamol was observed. Results: Mice liver injury model can be successfully established by gavage with ethionine or intraperitoneal injection of paracetamol. High and low-dose jujuba polysaccharide and the glucurolactone groups can significantly lower the ALT levels in serum and significantly improve pathological changes in liver injury model. Conclusion: Jujube polysaccharide had good protective effects on liver injury model mice induced by ethionine or paracetamol.

Object To preliminarily study the difference of proteins in the seed of spine date and the seed of Indian jujube. Methods Protein contents were measured with Kieldahl method. Amino acids constituting the protein were determined by means of acidolysis. The proteins were analyzed using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and capillary zone electrophoresis (CZE). Results Contents of proteins in the seed of spine date and the seed of Indian jujube were 36.13% and 41.58%, respectively. The content of valine and methionine in the seed of Indian jujube were significantly more than that in the seed of spine date remarkably, and the contents of other amino acids from the seed of spine date and seed of Indian jujube were similar. Their SDS-PAGE spectra showed that the seed of spine date contained a protein with molecular weight 39 800 and the seed of Indian jujube contained a protein with molecular weight 50 100 . In CZE spectra for the seed of spine date, there was a peak at the mobile time of 3.1 min and in CZE spectra from the seed of Indian jujube, there was a peak at the mobile time of 4.8 min. Conclusion Protein content in the seed of Indian jujube is more than that in the seed of spine date; The content of valine and methionine in the seed of Indian jujube is more than that in the seed of spine date remarkably. The spectra of SDS-PAGE and CZE for the seed of spine date are different from that for the seed of Indian jujube clearly.

OBJECTIVE:To determine Sarsasapogenin in Jujube seed concentrated pills by RP-HPLC-ELSD.METHODS:Separation of Sarsasapogenin was performed on Zorbax C18 column with methanol-water (90:10)as a mobile phase at a flow rate of 1.0mL?min-1.The temperature of the drift tube was 85℃and the air flow-rate was at 1.71mL?min-1.RESULTS:The linear range of Sarsasapogenin was 0.112 7~0.676 2mg?mL-1(r=0.998 3).The average recovery was 99.83%(RSD=0.93%).CONCLUSION:The method is simple,accurate and reproducible,and suitable for the quality control of Jujube seed concentrated pills.
